Estradiol, Progesterone falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the basic idea with Bijuva (estradiol and progesterone). A compound in Ceylon Leadwort called plumbagin can slow down a liver enzyme known as CYP1A2, which helps your body clear estrogen. In theory, blocking that enzyme could let estradiol build up a little higher than usual, which might add to typical estrogen-related effects like breast tenderness, bloating, nausea, headaches, or spotting.

I want to be honest about how soft this evidence is: it comes only from lab dish testing, not from real people, so we don't know if it matters in everyday life. Estradiol also clears through other pathways. Still, since hormone therapy is something to take seriously, I'd run any Ceylon Leadwort use past your pharmacist or doctor first.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Both the estradiol and progesterone in Bijuva are processed by a liver enzyme called CYP3A4. Ceylon Leadwort contains plumbagin, which in lab studies has been shown to slow down that same enzyme. If it does the same thing in the body, your hormone levels could climb a bit higher than expected. That might mean more of the usual side effects, like breast tenderness, bloating, nausea, headaches, mood changes, or spotting.

Here's the honest part though: this is only based on test-tube research, not real people, so it's a theoretical concern rather than a proven problem. Still, since hormones can be sensitive, I'd run it by your pharmacist or doctor before pairing the two, and please don't change your dose on your own.

Bijuva combines estradiol (an estrogen) with progesterone, and it's usually taken to ease menopause symptoms like hot flashes and night sweats. The concern here is that lab studies suggest Ceylon Leadwort root may have anti-estrogen activity, meaning it could work against the estrogen part of your therapy.

In plain terms, the worry is that the supplement might blunt how well your hormone therapy works, so symptoms like flushing or sweats could creep back. I want to be honest though: this is based only on test-tube and animal research, not on people, so we really don't know how strong the effect is in real life. Since this is hormone therapy, it's worth a quick chat with your pharmacist or doctor before combining the two.
