Estrogen falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the basic idea. Your body uses a liver enzyme called CYP1A2 to help break down estrogen. A compound in Ceylon Leadwort, plumbagin, has been shown in lab dish studies to slow that enzyme down. If that slowing happened in a real person, estrogen could clear out a little more slowly and build up, which might mean stronger effects and more side effects like breast tenderness, bloating, nausea, headaches, or mood changes.

Now the reassuring part: this is only theoretical. It comes from test-tube research, not from people, so we honestly don't know if it matters in real life. Still, since hormone therapy is something to take seriously, please check with your pharmacist or doctor before pairing these two rather than guessing.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

If you're taking estrogen for menopause symptoms, here's the concern with Ceylon Leadwort. In lab studies, an extract of this plant's root showed anti-estrogen activity, meaning it may push against the very effect your estrogen is supposed to provide. In theory, that could blunt how well your therapy works, so symptoms like hot flashes, vaginal dryness, or night sweats might creep back.

I want to be honest about the evidence though: this comes only from test-tube and animal research, not from people. So it's more of a flag to keep in mind than a proven problem. Still, before adding this supplement, please check with your pharmacist or doctor so you don't risk weakening a treatment that's working for you.
