Ethinyl Estradiol, Norgestimate falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the deal with your birth control pill (Ortho Tri-Cyclen and similar) and Ceylon Leadwort. A compound in this plant called plumbagin slowed down a liver enzyme (CYP1A2) in lab dishes. The estrogen part of your pill, ethinyl estradiol, is partly handled by that same enzyme. So in theory, if that pathway gets blocked, estrogen levels could nudge upward, which might mean more estrogen-type side effects like nausea, breast tenderness, headaches, or bloating.

The big caveat: this is only a test-tube finding, and it has never been reported in real people. So I wouldn't lose sleep over it. Still, because reliable birth control matters and we don't have good human data on this herb, I'd run it past your pharmacist or doctor before combining the two.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Your birth control pill (Ortho Tri-Cyclen and similar) contains hormones that your liver clears partly through an enzyme called CYP3A4. A compound in Ceylon Leadwort, plumbagin, has slowed down that same enzyme in lab dish experiments. In theory, if this enzyme works more slowly, the estrogen and progestin in your pill could build up a bit higher than usual, which might mean more of the typical hormone-related side effects like nausea, breast tenderness, headaches, or breakthrough spotting.

Here is the honest part: this is only test-tube evidence and has never actually been reported in people, so the real-world risk is likely small. Still, since Ceylon Leadwort itself can be irritating and isn't well studied, I'd run it by your pharmacist or doctor before combining the two.

Your birth control pill works by giving your body steady levels of estrogen (ethinyl estradiol) and a progestin (norgestimate) to prevent ovulation. Here's the thing about Ceylon Leadwort: in lab studies, a compound in its root seemed to push back against estrogen activity. In theory, that anti-estrogen effect could work against the hormones in your pill.

I want to be honest about how thin this evidence is. It comes only from test tube and animal research, not from people taking the pill, so we really don't know if it matters in real life. Still, since the worry would be weaker pill effects (and a small chance of breakthrough bleeding or reduced protection), I'd play it safe. Please check with your pharmacist or doctor before combining the two.
