Etonogestrel, Ethinyl Estradiol falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the honest picture with your NuvaRing (etonogestrel and ethinyl estradiol) and Ceylon Leadwort. A compound in this plant called plumbagin slowed down a liver enzyme (CYP1A2) in lab dishes. Ethinyl estradiol is partly handled by that enzyme, so in theory blocking it could let estrogen levels drift a little higher. That might mean more of the usual estrogen-type side effects, like nausea, breast tenderness, headaches, or bloating.

I want to be clear that this is only test-tube data and has never been seen in real people, so I wouldn't lose sleep over it. Still, since this plant isn't well studied and can be irritating in larger doses, please run it by your pharmacist or doctor before combining the two. Don't change how you use your ring on your own.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Your NuvaRing releases two hormones, etonogestrel and ethinyl estradiol, and your body clears them partly through a liver enzyme called CYP3A4. In lab dishes, the plumbagin in Ceylon Leadwort appears to slow that enzyme down. In theory, that could let these hormones build up a little higher than usual, which might mean more of the typical side effects like nausea, breast tenderness, headaches, mood changes, or breakthrough spotting.

I want to be honest with you though: this is only seen in test-tube studies, never reported in real people, so it's a soft, theoretical concern. These are not prodrugs, so the direction here is potential build-up, not loss of effect. Still, before pairing them, check in with your pharmacist or doctor.

Your NuvaRing slowly releases two hormones, etonogestrel (a progestin) and ethinyl estradiol (an estrogen), to prevent pregnancy. The concern here is that lab studies hint Ceylon Leadwort root may have anti-estrogenic activity, meaning it could theoretically work against the estrogen part of your ring.

I want to be honest about how thin this evidence is. It comes only from test-tube and animal research, not from real people using a contraceptive ring, so we truly don't know if it matters in everyday life. Still, since reliable birth control is the goal, I'd rather not gamble on a supplement that could blunt your hormones.
