Hydrocodone, Ibuprofen falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2D6 (CYP2D6) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2D6. Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), codeine, desipramine (Norpramin), flecainide (Tambocor), fluoxetine (Prozac), ondansetron (Zofran), tramadol (Ultram), and others.

Here's something interesting about this combo. Vicoprofen contains hydrocodone (an opioid for pain) plus ibuprofen. Hydrocodone is partly converted by an enzyme called CYP2D6 into hydromorphone, a stronger painkilling form. Ceylon Leadwort contains plumbagin, which has slowed down CYP2D6 in lab dishes. If that happened in people, it could mean less of that potent conversion, so your pain relief might feel weaker than expected.

I want to be honest though: this is only test-tube data, never confirmed in actual patients, so the real-world effect may be small or nothing at all. Still, if you take Vicoprofen and are considering this supplement, run it by your pharmacist or doctor first rather than guessing.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2C9 (CYP2C9) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2C9. Drugs that are metabolized by CYP2C9 include celecoxib (Celebrex), diclofenac (Voltaren), fluvastatin (Lescol), glipizide (Glucotrol), ibuprofen (Advil, Motrin), irbesartan (Avapro), losartan (Cozaar), phenytoin (Dilantin), piroxicam (Feldene), tamoxifen (Nolvadex), tolbutamide (Tolinase), torsemide (Demadex), and S-warfarin (Coumadin).

Vicoprofen pairs hydrocodone (an opioid) with ibuprofen (an anti-inflammatory). The ibuprofen part is cleared mainly by a liver enzyme called CYP2C9. In test-tube research, plumbagin from Ceylon Leadwort appears to slow that enzyme down. In theory, if ibuprofen clears more slowly, more could linger in your body, which might raise the odds of typical ibuprofen side effects like stomach upset, heartburn, ulcers or bleeding, and fluid retention.

I want to be honest about the strength here: this is only lab data, with nothing reported in actual people, so the real-world risk is likely small. Still, since this combo already carries some bleeding and stomach risk on its own, it is worth a quick chat with your pharmacist or doctor before adding the supplement.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Vicoprofen combines hydrocodone (an opioid) with ibuprofen (an anti-inflammatory). Your liver uses an enzyme called CYP3A4 to help process the hydrocodone part. A compound in Ceylon Leadwort, called plumbagin, has slowed that same enzyme in lab dish studies. If that held true in people, the hydrocodone could clear more slowly and build up a bit, which might mean stronger effects like extra drowsiness, dizziness, constipation, or slowed breathing.

Here's the honest part: this is only based on test-tube research, not real patients, so it's theoretical and far from certain. Still, since opioids carry real risks, I'd play it safe. Please check with your pharmacist or doctor before pairing concentrated Ceylon Leadwort supplements with this medication.

ANTICOAGULANT/ANTIPLATELET DRUGS

Hydrocodone, Ibuprofen also falls under ANTICOAGULANT/ANTIPLATELET DRUGS.

There is some concern that Ceylon leadwort might potentiate the effects of anticoagulant and antiplatelet drugs and possibly increase the risk of bleeding. Ceylon leadwort root extract has been shown to decrease platelet adhesion and prolong bleeding in animals. However, this effect has not yet been demonstrated in humans. Until more is known, use cautiously in patients taking anticoagulant or antiplatelet drugs. Some of these drugs include aspirin, clopidogrel (Plavix), dalteparin (Fragmin), enoxaparin (Lovenox), heparin, ticlopidine (Ticlid), warfarin (Coumadin), and others.

Here's the thing to keep in mind with Vicoprofen: the ibuprofen half is an NSAID, and NSAIDs already nudge up bleeding risk on their own by making platelets a little less sticky and by being hard on the stomach lining. Animal studies suggest Ceylon Leadwort may also thin the blood and lengthen bleeding time, so stacking the two could, in theory, add up to more bruising, nosebleeds, or stomach upset and bleeding.

I'll be honest, this concern comes from lab and animal research, not human studies, so it's far from proven. Still, since ibuprofen carries its own bleeding and ulcer risk, I'd play it safe. Please run this by your pharmacist or doctor before combining them, especially if you also take aspirin or a blood thinner.
