Levonorgestrel, Ethinyl Estradiol falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the gist with your birth control pill (the levonorgestrel and ethinyl estradiol combo). A compound in Ceylon Leadwort called plumbagin has been shown to slow down a liver enzyme called CYP1A2 in test-tube studies. Estrogens like the ethinyl estradiol in your pill are partly handled by that enzyme, so in theory blocking it could let estrogen levels drift a little higher. That might mean more of the usual estrogen-type side effects, such as nausea, breast tenderness, headaches, or bloating.

I want to be straight with you though: this is only lab and animal data, and it has never actually been reported in people. So it's more of a maybe than a real-world warning. Still, since reliable contraception matters, it's worth a quick word with your pharmacist or doctor before adding this supplement.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Your birth control pill (Nordette, Microgynon, and similar) contains ethinyl estradiol, an estrogen that your liver clears partly through an enzyme called CYP3A4. In lab tests, plumbagin from Ceylon Leadwort appears to slow that same enzyme down. Your pill, a combination of levonorgestrel and ethinyl estradiol, is a birth control pill that works by delivering steady estrogen and progestin to prevent pregnancy. The concern here is that lab studies hint Ceylon Leadwort root may have anti-estrogen activity, meaning it could in theory work against part of what your pill is doing.

I want to be honest about how thin this evidence is. It comes only from test-tube and animal research, not from people taking the pill, so we really do not know if it matters in real life. Still, since reliable birth control is something you do not want to gamble on, I would not add a concentrated Leadwort supplement without first checking with your doctor or pharmacist.
