Why this applies to Lurasidone

Lurasidone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Here's the gist for Latuda (lurasidone). Your body relies heavily on an enzyme called CYP3A4 to clear this medication out. In lab studies, plumbagin (the main active compound in Ceylon Leadwort) appears to block that same enzyme. If that blocking happened in a real person, lurasidone could build up higher than intended, which might mean more drowsiness, dizziness, restlessness, muscle stiffness, or trouble with movements.

The important caveat: this has only been seen in test tubes, not in actual people, so it's theoretical for now. Lurasidone is also fussy about CYP3A4 already (grapefruit is off-limits with it), so I'd lean cautious. Please run it by your pharmacist or prescriber before combining the two.

Ceylon Leadwort (Plumbago zeylanica) is a plant long used in Ayurvedic and other traditional medicine systems, mainly for digestion, skin problems, and pain. Solid human studies are lacking, and the plant contains plumbagin, a compound that can irritate skin and tissues and may be unsafe in larger amounts. What Is Lurasidone Used For?

Brand name: Latuda

Lurasidone hydrochloride tablets are approved to treat schizophrenia in adults and adolescents aged 13 to 17, and to treat depressive episodes associated with bipolar I disorder (bipolar depression) in adults and children aged 10 to 17. Latuda is one of the brand names under which lurasidone is available as a tablet.

For bipolar depression, lurasidone can be used on its own (monotherapy) in adults and pediatric patients aged 10 to 17, or used alongside lithium or valproate (adjunctive therapy) in adults.
