Segesterone Acetate, Ethinyl Estradiol falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the gist with Annovera (the segesterone acetate and ethinyl estradiol vaginal ring) and Ceylon Leadwort. A compound in this plant called plumbagin has slowed down a liver enzyme called CYP1A2 in test-tube studies. Ethinyl estradiol is partly handled by that enzyme, so in theory the supplement could nudge estrogen levels a bit higher. If that happened, you might notice more typical estrogen-related effects like nausea, breast tenderness, headaches, or breakthrough spotting.

I want to be honest though: this is lab-only evidence and hasn't been seen in real people, so I wouldn't lose sleep over it. Plumbagin can also irritate tissues and isn't well studied in humans. Before adding Ceylon Leadwort, run it past your pharmacist or doctor first.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Annovera is a vaginal ring that releases two hormones, segesterone acetate and ethinyl estradiol, to prevent pregnancy. Both of these hormones are partly cleared by a liver enzyme called CYP3A4. In lab dish studies (not in actual people), plumbagin from Ceylon Leadwort appeared to slow that same enzyme down. If that held true in the body, your hormone levels could drift a little higher, which might mean more of the usual estrogen-related effects like nausea, breast tenderness, headaches, or spotting.

Here's the reassuring part: this is only a theoretical concern based on test-tube work, and it has never been reported in real patients. I wouldn't lose sleep over it, but since Annovera is something you rely on, it's worth a quick chat with your pharmacist or doctor before adding this supplement.

Annovera is a vaginal ring that delivers two hormones, segesterone (a progestin) and ethinyl estradiol (an estrogen), to prevent pregnancy. The concern here is that, in lab studies only, Ceylon Leadwort root has shown anti-estrogenic activity. In plain terms, that means it could theoretically push against the estrogen part of your ring and possibly weaken how well it works.

I want to be honest about how thin this evidence is. We're talking about test-tube and animal research, not real people, so we don't actually know if this matters in everyday use. Still, because reliable birth control is the whole point, I'd play it safe. Please check with your doctor or pharmacist before combining the two, and don't stop your ring on your own.
