Why this applies to Celecoxib, Tramadol

Celecoxib, Tramadol falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Lentinan interacts with.

Laboratory research suggests that lentinan might increase the activity of cytochrome P450 2D6 (CYP2D6). So far, this interaction has not been reported in humans. However, watch for a decrease in the levels of drugs metabolized by CYP2D6 in patients taking lentinan.
Some drugs metabolized by CYP2D6 include tricyclic antidepressants such as imipramine (Tofranil) and amitriptyline (Elavil); antipsychotics such as haloperidol (Haldol), risperidone (Risperdal), and chlorpromazine (Thorazine); beta-blockers such as propranolol (Inderal), metoprolol (Lopressor, Toprol XL), and carvedilol (Coreg); tamoxifen (Nolvadex); and others.

Seglentis combines two medicines. The piece that matters here is tramadol, which is actually a prodrug. Your liver uses an enzyme called CYP2D6 to turn tramadol into its stronger, pain-relieving form. Lab studies hint that lentinan might speed up that enzyme. If that happened, tramadol could be activated faster, which sounds helpful but can mean a bit more sedation, dizziness, nausea, or constipation. The celecoxib half is barely affected by this pathway, so it isn't the main concern.

Here's the honest part: this is only test-tube and animal data, never confirmed in real people, so I wouldn't lose sleep over it. Still, if you're using oral lentinan and notice any changes in how you feel, please check in with your pharmacist or doctor before adjusting anything.

Laboratory research suggests that lentinan might increase the activity of cytochrome P450 3A4 (CYP3A4). So far, this interaction has not been reported in humans. However, watch for a decrease in the levels of drugs metabolized by CYP3A4 in patients taking lentinan.
Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others.

Seglentis combines two medicines: celecoxib, an anti-inflammatory pain reliever, and tramadol, an opioid-type pain medicine. Both rely partly on the liver enzyme CYP3A4 to be processed. The lab research we have suggests lentinan might speed that enzyme up, which could mean your body clears these drugs a little faster and you get less pain relief than expected.

One twist worth noting: tramadol is a prodrug, so it needs liver enzymes to turn it into its active painkilling form. The picture there is genuinely mixed, which is another reason not to read too much into this. Honestly, this is only test-tube and animal data, never confirmed in people, so I wouldn't lose sleep over it. Still, if you take both, mention it to your pharmacist or doctor and watch whether your pain control changes.
