Glutamine is generally well tolerated by healthy adults. Common side effects from oral doses include belching, bloating, constipation, diarrhea, flatulence, stomach pain, nausea, vomiting, headache, cough, and musculoskeletal pain.

Dizziness and headache have also been reported; one person's dizziness resolved when the dose was lowered. There are two reported cases of mania or hypomania in people with bipolar disorder taking up to 4 grams daily, though this is rare.
