Here's the gist: your body turns adrafinil into modafinil, and modafinil can put the brakes on a liver enzyme called CYP2C19. That same enzyme is one of the main ways your body clears omeprazole. If it slows down, more omeprazole can linger in your system, nudging its levels and effects upward.

The good news is omeprazole is a pretty forgiving medicine, so a little extra usually isn't dangerous. You might notice more of its typical effects, like headache, nausea, gas, or loose stools. Keep in mind this is a theoretical concern based on lab studies, not real-world reports, so I wouldn't lose sleep over it. Still, since adrafinil is unregulated and hard on the liver, please run it by your pharmacist or doctor first.

Here's the gist for these two together. Once your liver turns adrafinil into modafinil, that modafinil can gently speed up a liver enzyme called CYP3A4. Omeprazole is partly handled by that same enzyme, so in theory your body might clear it a little faster. If that happens, omeprazole could become slightly less effective, meaning heartburn, acid reflux, or stomach discomfort might creep back in.

I want to be honest about the strength here: this is a theoretical concern based on how the drugs work on paper, not something well documented in real patients. The effect is also described as weak. Still, if you take omeprazole for GERD or an ulcer and notice symptoms returning, mention the adrafinil to your pharmacist or doctor before changing anything.

About Omeprazole

What Is Omeprazole Used For?

Brand names: Prilosec, Zegerid, Losec

Omeprazole is used to treat a range of conditions where stomach acid causes problems. These include heartburn and acid reflux (GERD), erosive esophagitis (damage to the esophagus from acid), and stomach or duodenal ulcers. It can also be used alongside antibiotics to help get rid of an H. pylori infection — a bacteria that causes ulcers.

In more serious cases, omeprazole is used to manage conditions where the stomach produces far too much acid, such as pathologic hypersecretory conditions (rare medical disorders that cause extreme acid overproduction). It is approved for adults and children one year of age and older for GERD and erosive esophagitis.
