Entrectinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the gist for Rozlytrek (entrectinib): your body relies on a liver enzyme called CYP3A4 to clear this medicine out. Ayahuasca contains beta-carboline compounds that, at least in lab dishes, can slow that same enzyme down. If that slowdown happened in real life, entrectinib could build up in your blood and hit harder than intended.

For entrectinib, more drug in your system can mean more dizziness, fatigue, swelling, constipation, changes in heart rhythm, or trouble with balance and thinking. That said, this is theoretical, based only on test-tube work and never confirmed in people. Even so, since you're treating cancer with a drug that needs careful dosing, please don't combine these without checking with your oncologist or pharmacist first.

About Entrectinib

What Is Entrectinib Used For?

Brand name: Rozlytrek

Rozlytrek (entrectinib) is used to treat adult patients with ROS1-positive metastatic non-small cell lung cancer (NSCLC), as confirmed by an FDA-approved test. It is also approved for adult and pediatric patients older than 1 month of age who have solid tumors — any cancer type — that carry an NTRK gene fusion detected by an FDA-approved test, provided the tumor has no known acquired resistance mutation, is metastatic or cannot be surgically removed without severe harm, and has progressed after prior treatment or has no satisfactory alternative therapy.

The NTRK gene fusion approval in solid tumors was granted under accelerated approval, meaning it is based on tumor response rates. Continued approval may depend on results from confirmatory clinical trials.
