Aripiprazole Lauroxil falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2D6 (CYP2D6) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2D6. Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), codeine, desipramine (Norpramin), flecainide (Tambocor), fluoxetine (Prozac), ondansetron (Zofran), tramadol (Ultram), and others.

Aristada (aripiprazole lauroxil) is a long-acting injection your body partly clears using a liver enzyme called CYP2D6. A compound in Ceylon Leadwort, plumbagin, has been shown to slow down that same enzyme, but so far only in lab dish testing, not in real people. In theory, blocking CYP2D6 could let aripiprazole build up a bit more than usual, which might mean stronger side effects like drowsiness, restlessness, dizziness, tremor, or trouble with movements.

Honestly, the evidence here is weak and unproven in humans, so I wouldn't lose sleep over it. Still, since this is a long-acting shot you can't easily take back, it's worth mentioning Ceylon Leadwort to your pharmacist or prescriber before combining them.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Here's the gist for Aristada (aripiprazole lauroxil): your body relies partly on an enzyme called CYP3A4 to break this medication down. In lab studies, a compound in Ceylon Leadwort called plumbagin appears to slow that same enzyme. In theory, that could let aripiprazole build up a bit more than expected. Since this is a long-acting injection you receive at intervals, higher levels could mean more of its usual side effects, things like drowsiness, restlessness or inability to sit still, dizziness, tremor, or feeling unsteady.

Ceylon Leadwort (Plumbago zeylanica) is a plant long used in Ayurvedic and other traditional medicine systems, mainly for digestion, skin problems, and pain. Solid human studies are lacking, and the plant contains plumbagin, a compound that can irritate skin and tissues and may be unsafe in larger amounts. Aripiprazole injection is used to treat schizophrenia and to provide maintenance treatment for bipolar I disorder in adults. Different injectable products carry these approvals.

Abilify Asimtufii is approved for the treatment of schizophrenia in adults and as maintenance monotherapy for bipolar I disorder in adults. Aristada is approved for the treatment of schizophrenia in adults, with Aristada Initio used specifically to help start Aristada therapy.
