Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2D6. Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), codeine, desipramine (Norpramin), flecainide (Tambocor), fluoxetine (Prozac), ondansetron (Zofran), tramadol (Ultram), and others.

Your body relies on a liver enzyme called CYP2D6 to help break down imipramine. Plumbagin, the active compound in Ceylon Leadwort, has been shown in lab dishes to slow that enzyme down. If that same effect held true in people, your imipramine could clear out more slowly and build up to higher levels than usual. That means a better chance of side effects you'd expect from this tricyclic, like drowsiness, dry mouth, constipation, blurry vision, dizziness on standing, or a faster heartbeat.

Here's the gist for imipramine (Tofranil). Your liver uses an enzyme called CYP1A2 to help clear this antidepressant out of your system. A compound in Ceylon Leadwort, plumbagin, has been shown to slow that enzyme down. Imipramine is active as-is (not a prodrug), so if its breakdown slows, more of it can build up in your blood than expected.

In practice, that could mean stronger tricyclic side effects: dry mouth, constipation, blurry vision, dizziness, drowsiness, or a faster heartbeat. The important caveat: this is only seen in lab studies so far, never confirmed in real people, so it stays theoretical. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2C9. Drugs that are metabolized by CYP2C9 include celecoxib (Celebrex), diclofenac (Voltaren), fluvastatin (Lescol), glipizide (Glucotrol), ibuprofen (Advil, Motrin), irbesartan (Avapro), losartan (Cozaar), phenytoin (Dilantin), piroxicam (Feldene), tamoxifen (Nolvadex), tolbutamide (Tolinase), torsemide (Demadex), and S-warfarin (Coumadin).

Ceylon Leadwort (Plumbago zeylanica) is a plant long used in Ayurvedic and other traditional medicine systems, mainly for digestion, skin problems, and pain. Solid human studies are lacking, and the plant contains plumbagin, a compound that can irritate skin and tissues and may be unsafe in larger amounts. What Is Imipramine Used For?

Brand names: Tofranil, Tofranil PM

Imipramine is used to relieve symptoms of depression. Both Imipramine Hydrochloride tablets and Imipramine Pamoate capsules are approved for this use. Endogenous depression — the kind that seems to arise from within rather than a clear outside cause — tends to respond especially well. It may take one to three weeks before you notice the full benefit.

Imipramine Hydrochloride tablets are also approved as a short-term, add-on treatment to help reduce nighttime bedwetting (nocturnal enuresis) in children aged 6 and older, after a doctor has ruled out physical causes.
