Olanzapine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2D6 (CYP2D6) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2D6. Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), codeine, desipramine (Norpramin), flecainide (Tambocor), fluoxetine (Prozac), ondansetron (Zofran), tramadol (Ultram), and others.

Here's the gist for olanzapine (Zyprexa): your body partly clears it using a liver enzyme called CYP2D6. A compound in Ceylon Leadwort, plumbagin, has slowed down that enzyme in lab tests. If that same effect happened in people, olanzapine could clear out a little slower and build up in your system.

Olanzapine isn't a prodrug, so more drug hanging around generally means stronger effects and more side effects, things like heavy drowsiness, dizziness, weight gain, or feeling sluggish. The reassuring part: this is only test-tube evidence, never actually reported in humans, and CYP2D6 is a minor pathway for olanzapine. So the real-world risk is likely small. Still, since this affects your mental health treatment, run it by your pharmacist or doctor before combining them.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the gist: your body uses a liver enzyme called CYP1A2 to clear olanzapine out of your system. A compound in Ceylon Leadwort, plumbagin, has been shown in lab dish studies to slow that same enzyme down. If it does the same thing in people, olanzapine could clear out more slowly and build up a bit higher than usual.

What might that feel like? Mostly more of olanzapine's usual effects, like feeling drowsy or sedated, dizzy when standing up, dry mouth, or weight gain. The important part: this is only theoretical right now, based on test-tube work and never reported in actual patients, so I wouldn't lose sleep over it. Still, since this is a medication for serious conditions, please run it by your pharmacist or doctor before adding the supplement.

Ceylon Leadwort (Plumbago zeylanica) is a plant long used in Ayurvedic and other traditional medicine systems, mainly for digestion, skin problems, and pain. Solid human studies are lacking, and the plant contains plumbagin, a compound that can irritate skin and tissues and may be unsafe in larger amounts. About Olanzapine

What Is Olanzapine Used For?

Brand name: Zyprexa

Olanzapine treats schizophrenia and the manic or mixed episodes of bipolar I disorder in both adults and adolescents (ages 13–17). It can also be used alongside valproate or lithium to treat manic or mixed episodes in adults. When combined with fluoxetine, olanzapine tablets and orally disintegrating tablets are used to treat depressive episodes associated with bipolar I disorder and treatment-resistant depression in adults.

Olanzapine intramuscular injection (short-acting) is used by healthcare providers to quickly calm acute agitation in adults with schizophrenia or bipolar I mania. Zyprexa Relprevv — a separate, long-acting intramuscular formulation available only through a restricted program — is indicated specifically for the treatment of schizophrenia in adults.
