Tisotumab Vedotin-Tftv falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Here's the gist with Tivdak (tisotumab vedotin-tftv) and Ceylon Leadwort. Tivdak is an antibody-drug conjugate, meaning the antibody delivers a potent cell-killing payload (called MMAE) to cancer cells. That payload is partly broken down by a liver enzyme called CYP3A4. Plumbagin, a compound in Ceylon Leadwort, has slowed this enzyme down in lab dishes. In theory, that could let more of the active payload build up, which might mean more side effects like nausea, low blood counts, fatigue, hair loss, nosebleeds, or eye irritation.

I'll be straight with you: this is only test-tube data and has not been seen in real patients, so don't panic. But since Tivdak is serious cancer chemotherapy, please loop in your oncologist or pharmacist before adding any supplement.
