Venlafaxine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2D6 (CYP2D6) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2D6. Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), codeine, desipramine (Norpramin), flecainide (Tambocor), fluoxetine (Prozac), ondansetron (Zofran), tramadol (Ultram), and others.

Here's the gist for venlafaxine (Effexor): your body relies on a liver enzyme called CYP2D6 to help break it down. In lab studies, plumbagin (a compound in Ceylon Leadwort) appears to block that enzyme. If that happened in real life, venlafaxine could clear out of your body more slowly and build up a bit, which might mean stronger effects and more side effects like nausea, jitteriness, trouble sleeping, a faster heartbeat, higher blood pressure, or sweating.

I want to be honest about the evidence, though: this has only been seen in a test tube, never in actual people, so it's theoretical for now. Still, before pairing Ceylon Leadwort with venlafaxine, it's worth a quick chat with your pharmacist or doctor.

Venlafaxine is used to treat major depressive disorder (MDD) across all available formulations. Extended-release capsules (including Effexor XR) are also approved for generalized anxiety disorder (GAD), social anxiety disorder (SAD), and panic disorder (PD).

Extended-release tablets are approved for major depressive disorder and social anxiety disorder. Immediate-release venlafaxine tablets are indicated for major depressive disorder only.
