Chanca Piedra + Medroxyprogesterone
Medroxyprogesterone brand names: Provera, Amen, Depo-Provera, Depo-subQ Provera 104.
Chanca Piedra has a moderate interaction with Medroxyprogesterone. Theoretically, use of chanca piedra might increase the levels and clinical effects of CYP3A4 substrates.

Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
Why this applies to Medroxyprogesterone
Medroxyprogesterone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Chanca Piedra interacts with.
In vitro research shows that chanca piedra extract inhibits CYP3A4. Theoretically, chanca piedra might increase the levels of CYP3A4 substrates. This interaction has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the short version for Provera (medroxyprogesterone). Your body relies on a liver enzyme called CYP3A4 to clear this hormone out of your system. In lab dish studies, chanca piedra appears to slow that same enzyme down. If that held true in people, your medroxyprogesterone could clear a little more slowly and build up, which might mean more of its usual effects like bloating, breast tenderness, mood changes, headache, or spotting.
The important thing to know: this is theory from test-tube research only. It has not actually been seen in real patients, so I wouldn't lose sleep over it. Still, since you're combining a hormone with an herb, it's worth a quick chat with your pharmacist or doctor before starting chanca piedra.

From our pharmacist’s overview on the Medroxyprogesterone page
“Medroxyprogesterone (Provera, Depo-Provera) only works if its blood level holds steady, and 121 of the 139 supplements and herbs on file can shift that level through a single liver enzyme, CYP3A4. That makes this a list to respect, with three entries rated major.”

About Chanca Piedra
What is Chanca Piedra?
- Kidney stones
- Gallstones
- Liver support
- Urinary tract health
- Digestive complaints
Chanca piedra is a tropical herb traditionally used as a 'stone breaker' for kidney and gallstones, and for liver and urinary health. Human evidence for these uses is limited and mostly small or preliminary, so it should not replace proven treatments. Talk with your pharmacist or doctor before using it, especially if you have a medical condition or take medications.
About Medroxyprogesterone
What Is Medroxyprogesterone Used For?
Brand names: Provera, Amen, Depo-Provera, Depo-subQ Provera 104
Medroxyprogesterone is used for different conditions depending on the product form. The injectable forms — including Depo-Provera and intramuscular medroxyprogesterone acetate injectable suspension — are used to prevent pregnancy. Depo-subQ Provera 104 (the subcutaneous injection) is also approved to manage pain associated with endometriosis.
The oral tablets — sold under the brand Provera and as generic medroxyprogesterone acetate tablets — treat secondary amenorrhea (periods that have stopped) and abnormal uterine bleeding caused by hormonal imbalance. The tablets are also used to prevent endometrial hyperplasia (abnormal thickening of the uterine lining) in postmenopausal women with a uterus who are taking daily conjugated estrogens.
