Cyclobenzaprine falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Chanca Piedra interacts with.
In vitro research shows that chanca piedra extract increases CYP1A2 activity. Theoretically, chanca piedra might increase metabolism of CYP1A2 substrates and lower serum concentrations. This interaction has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for these two. Your body relies partly on an enzyme called CYP1A2 to clear cyclobenzaprine, and lab studies suggest chanca piedra may speed that enzyme up. If that happened in real life, your system could break down the muscle relaxant a bit faster, which might leave you with less relief from the muscle pain, tightness, or spasms you're trying to ease. Cyclobenzaprine is the active drug itself, not a prodrug, so faster clearance means a weaker effect, not a stronger one.
The big caveat: this is only test-tube evidence and has never been reported in actual people, so it may not matter at all. Still, if you notice the medication isn't working as well, check in with your pharmacist or doctor before changing anything.

In vitro research shows that chanca piedra extract inhibits CYP3A4. Theoretically, chanca piedra might increase the levels of CYP3A4 substrates. This interaction has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the short version: your body relies partly on an enzyme called CYP3A4 to clear cyclobenzaprine out of your system. In lab studies (test tube only, not in actual people), chanca piedra appears to slow that enzyme down. If that same effect happened in your body, cyclobenzaprine could linger longer and build up a bit, which might make its usual side effects stronger.
For cyclobenzaprine that could mean more drowsiness, dizziness, dry mouth, blurry vision, or a groggy, foggy feeling. The good news is this is theoretical and hasn't been reported in real patients, so I wouldn't panic. Still, since both can make you sleepy, it's worth a quick chat with your pharmacist or doctor before combining them, especially before driving.

What Is Cyclobenzaprine Used For?
Brand names: Cyclobenzaprine, Flexeril, Cycloprine
Cyclobenzaprine tablets and extended-release capsules are used alongside rest and physical therapy to relieve muscle spasm caused by acute, painful musculoskeletal conditions — easing pain, tenderness, and limited movement. This use is intended for short-term treatment only (up to 2–3 weeks).
Tonmya, the sublingual (under-the-tongue) form of cyclobenzaprine, is separately indicated for the treatment of fibromyalgia in adults. It is not the same as the oral tablet or capsule formulations.
