Hordenine is structurally similar to N-methyltyramine and synephrine, constituents in bitter orange known to have stimulant properties. Theoretically, taking hordenine with drugs with stimulant properties might increase the risk of hypertension and other adverse cardiovascular effects.
Some of these drugs include amphetamine, caffeine, methylphenidate, pseudoephedrine, and many others.

Here's the simple version: amphetamine (like Dyanavel XR or Mydayis) is already a strong stimulant. It revs up your body's fight-or-flight system, which can raise your heart rate and blood pressure. Hordenine appears to nudge those same pathways, possibly bumping up norepinephrine. So stacking the two could pile stimulant on top of stimulant, and you might notice a faster or pounding heartbeat, higher blood pressure, jitteriness, or trouble sleeping.

I want to be honest about the evidence, though. This concern is theoretical, based on how hordenine looks and behaves in lab studies, not on real-world cases. Amphetamine is not a prodrug, so we're talking about added effect, not less. Still, since hordenine isn't well studied, please check with your pharmacist or doctor before combining them.

Amphetamine also falls under MONOAMINE OXIDASE INHIBITORS (MAOIs).

Hordenine is structurally similar to tyramine In vitro research shows that hordenine is a selective substrate for monoamine oxidase-B in the liver. Theoretically, concomitant use of hordenine with MAOIs might increase blood pressure, potentially leading to a hypertensive crisis.
Some MAOIs include isocarboxazid (Marplan), phenelzine (Nardil), selegiline (Eldepryl, Emsam, Zelapar), and tranylcypromine (Parnate).

Here's the thing about pairing hordenine with your amphetamine: both lean on the same stress chemicals, like norepinephrine, that rev up your body. Amphetamine already pushes those messengers hard, and hordenine is a mild stimulant that may nudge them in the same direction. So stacking them could, in theory, add up to a bigger jolt than you bargained for, think racing heart, jitteriness, trouble sleeping, and most importantly a spike in blood pressure.

I want to be honest with you, though. This warning comes from lab work and the fact that hordenine looks chemically a lot like tyramine. We don't have real human studies showing it happens, so it's more of a caution than a proven danger. Still, with a heart-and-blood-pressure medication like this, I'd run it by your pharmacist or doctor before combining them.

Amphetamine also falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES.

Hordenine weakly inhibits cytochrome P450 2D6 (CYP2D6) enzymes in vitro. Theoretically, hordenine might increase the levels of CYP2D6 substrates.
Some of drugs that are CYP2D6 substrates include amitriptyline (Elavil), clozapine (Clozaril), codeine, desipramine (Norpramin), donepezil (Aricept), fentanyl (Duragesic), flecainide (Tambocor), fluoxetine (Prozac), meperidine (Demerol), methadone (Dolophine), metoprolol (Lopressor, Toprol XL), olanzapine (Zyprexa), ondansetron (Zofran), tramadol (Ultram), trazodone (Desyrel), and others.

Here's the gist with this pairing. Your body uses an enzyme called CYP2D6 to help clear amphetamine. In lab dish studies (not in people), hordenine appears to be a weak blocker of that enzyme. If that effect carried over into real life, amphetamine could clear a bit slower and build up slightly, which might mean more of its usual effects: a faster heartbeat, higher blood pressure, jitteriness, trouble sleeping, or a reduced appetite.

That said, I don't want to overstate this. The evidence is theoretical and based only on test-tube work, and the interaction is rated minor. On top of that, hordenine is itself a stimulant, so stacking two stimulants is the bigger common-sense concern. Before combining them, please check with your pharmacist or prescriber.

What Is Amphetamine Used For?

Brand names: Dyanavel XR, Adensys XR-ODT, Mydayis, Adzenys ER

Amphetamine treats Attention Deficit Hyperactivity Disorder (ADHD) in patients 6 years and older. Adzenys XR-ODT, Dyanavel XR, and the amphetamine extended-release orally disintegrating tablet formulations are specifically approved for ADHD in this age group.

Amphetamine Sulfate tablets and Evekeo (amphetamine sulfate tablets) are also approved for narcolepsy (a sleep disorder causing uncontrollable daytime sleepiness) and as a short-term aid in a calorie-restricted weight-loss program for exogenous obesity (weight gain from external factors like overeating) in patients who have not responded to other approaches.
