Hordenine + Amitriptyline
Amitriptyline brand name: Elavil.
Hordenine has a minor interaction with Amitriptyline. Hordenine weakly inhibits cytochrome P450 2D6 (CYP2D6) enzymes in vitro.

Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES
Why this applies to Amitriptyline
Amitriptyline falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Hordenine interacts with.
Some of drugs that are CYP2D6 substrates include amitriptyline (Elavil), clozapine (Clozaril), codeine, desipramine (Norpramin), donepezil (Aricept), fentanyl (Duragesic), flecainide (Tambocor), fluoxetine (Prozac), meperidine (Demerol), methadone (Dolophine), metoprolol (Lopressor, Toprol XL), olanzapine (Zyprexa), ondansetron (Zofran), tramadol (Ultram), trazodone (Desyrel), and others.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with hordenine and your amitriptyline (Elavil). Your liver uses an enzyme called CYP2D6 to help break amitriptyline down and clear it out. In test-tube studies, hordenine appears to mildly slow that same enzyme. In theory, that could let a bit more amitriptyline build up in your system, which might nudge up its usual side effects like drowsiness, dry mouth, constipation, blurred vision, or a faster heartbeat.
I want to be honest about the strength here: this is rated minor and comes only from lab data, not from real people, so it's a weak, theoretical concern. Still, hordenine is a stimulant with little human safety data, and amitriptyline already affects the heart. I'd run this combo by your pharmacist or doctor before adding it.

Every interaction above is graded three ways — here’s the key to each label.
Clinically significant. Avoid the combination, or use only under close professional supervision.
Usually worth avoiding or managing — may need monitoring, a dose change, or timing apart.
Generally not serious. Be aware of it and mention it to your pharmacist.
- LikelyWell-controlled human studies have demonstrated the likely existence of this interaction.
- ProbableInteraction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports.
- PossibleInteraction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
- UnlikelyInteraction has been demonstrated in animal or in lab research but has been shown not to occur in humans.

“Amitriptyline (Elavil), an older antidepressant now often prescribed for nerve pain, migraines, and sleep, is one of the drugs I genuinely want patients to be careful with, and 17 of the 296 supplement and herbal ingredients on this list carry a major rating. About Hordenine
What is Hordenine?
- Energy and stimulation
- Athletic and workout performance
- Weight loss/fat burning
- Focus and mood
- Appetite control
Hordenine is a natural alkaloid found in barley and some cacti that is marketed as a stimulant for energy, focus, and fat loss, but solid human evidence for these benefits is lacking. Its safety in people has not been well studied, so caution is warranted, and it should be avoided in pregnancy, breastfeeding, and by people with heart conditions.
About Amitriptyline
What Is Amitriptyline Used For?
Brand name: Elavil
Amitriptyline HCL and Amitriptyline Hydrochloride tablets are used to relieve symptoms of depression. Depression that originates from within the body (called endogenous depression) tends to respond better than other types of depression.

Amitriptyline Is Linked to Lower Levels of 3 Nutrients
Natural Medicines records Amitriptyline as a medicine that can lower these. This is not an interaction with Hordenine — it is a separate effect of the medicine, and it is a reason some people are advised to supplement rather than to avoid one:
