Why this applies to Doxepin

Doxepin falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Hordenine interacts with.

Hordenine weakly inhibits cytochrome P450 2D6 (CYP2D6) enzymes in vitro. Theoretically, hordenine might increase the levels of CYP2D6 substrates.
Some of drugs that are CYP2D6 substrates include amitriptyline (Elavil), clozapine (Clozaril), codeine, desipramine (Norpramin), donepezil (Aricept), fentanyl (Duragesic), flecainide (Tambocor), fluoxetine (Prozac), meperidine (Demerol), methadone (Dolophine), metoprolol (Lopressor, Toprol XL), olanzapine (Zyprexa), ondansetron (Zofran), tramadol (Ultram), trazodone (Desyrel), and others.

Here's the gist for doxepin: your body relies partly on an enzyme called CYP2D6 to clear it out. Hordenine has been shown, but only in lab dish studies, to weakly block that same enzyme. In theory, that could let doxepin build up a bit higher than usual. Doxepin isn't a prodrug, so more drug hanging around means a stronger effect, not a weaker one.

If that happened, you might feel extra drowsy or groggy in the morning, dry mouth, dizziness, or constipation. Honestly though, this concern rests on weak test-tube evidence and is rated minor, so I wouldn't lose sleep over it. Still, since hordenine is a stimulant and barely studied in people, it's worth a quick chat with your pharmacist or doctor before pairing the two.
