These two interact in 2 distinct ways. Each one is explained separately below.
Why Are There 2 Interactions?
Hordenine interacts with whole categories of medication. Chlorpheniramine, Hydrocodone, Pseudoephedrine falls into 2 of those categories, so it can interact with Hordenine in 2 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).
STIMULANT DRUGS
1 of 2Why this applies to Chlorpheniramine, Hydrocodone, Pseudoephedrine
Chlorpheniramine, Hydrocodone, Pseudoephedrine falls under STIMULANT DRUGS — the group Hordenine interacts with.
Some of these drugs include amphetamine, caffeine, methylphenidate, pseudoephedrine, and many others.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
The piece of this combination product to focus on is the pseudoephedrine, which is a decongestant that already nudges up your heart rate and blood pressure. Hordenine is marketed as a stimulant and looks chemically similar to compounds that rev up your 'fight-or-flight' signals. Stacking the two could, in theory, add together and push things like a racing heartbeat, jitteriness, higher blood pressure, trouble sleeping, or headache.
I'll be honest with you: this concern is theoretical, based on how these ingredients behave rather than real-world reports, so I don't want to alarm you. Still, since hordenine hasn't been well studied in people and isn't recommended for anyone with heart issues, it's worth running this combo past your pharmacist or doctor before mixing them.

CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES
2 of 2Chlorpheniramine, Hydrocodone, Pseudoephedrine also falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES.
Some of drugs that are CYP2D6 substrates include amitriptyline (Elavil), clozapine (Clozaril), codeine, desipramine (Norpramin), donepezil (Aricept), fentanyl (Duragesic), flecainide (Tambocor), fluoxetine (Prozac), meperidine (Demerol), methadone (Dolophine), metoprolol (Lopressor, Toprol XL), olanzapine (Zyprexa), ondansetron (Zofran), tramadol (Ultram), trazodone (Desyrel), and others.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the wrinkle with this combo product: the hydrocodone piece is a bit like codeine. It relies on a liver enzyme called CYP2D6 to turn into its more active, stronger painkilling form. Hordenine may weakly block that same enzyme. In theory, that could mean slightly less of the active form is made, so you might get a little less pain relief rather than more. The decongestant (pseudoephedrine) is mostly cleared elsewhere, so it's less affected.
One thing worth flagging separately: hordenine is a mild stimulant, and pseudoephedrine is too. Stacking two stimulants could bump up jitteriness, a racing heart, or higher blood pressure. The enzyme part is only lab-based and rated minor, so don't panic, but please run this past your pharmacist before pairing them.

From our pharmacist’s overview on the Chlorpheniramine, Hydrocodone, Pseudoephedrine page
“Tussend combines three drugs in one cough medicine, the opioid hydrocodone, the antihistamine chlorpheniramine, and the decongestant pseudoephedrine, so supplements can collide with it from several directions. - Energy and stimulation
- Athletic and workout performance
- Weight loss/fat burning
- Focus and mood
- Appetite control
Hordenine is a natural alkaloid found in barley and some cacti that is marketed as a stimulant for energy, focus, and fat loss, but solid human evidence for these benefits is lacking. Its safety in people has not been well studied, so caution is warranted, and it should be avoided in pregnancy, breastfeeding, and by people with heart conditions.
