Here is the interaction we found between these two, with the evidence behind it.
CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES
Why this applies to Acetaminophen, Aspirin
Acetaminophen, Aspirin falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Lentinan interacts with.
Some drugs metabolized by CYP1A2 include amitriptyline (Elavil), haloperidol (Haldol), ondansetron (Zofran), propranolol (Inderal), theophylline (Theo-Dur, others), verapamil (Calan, Isoptin, others), and others.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with this combo product (acetaminophen plus aspirin). The concern on file is that lentinan, a shiitake mushroom beta-glucan, might speed up a liver enzyme called CYP1A2. In theory, faster enzyme activity could mean a drug gets cleared a bit quicker. But honestly, this is based only on lab and animal data, not on real people, so I wouldn't lose sleep over it.
One thing worth a mention: acetaminophen's liver breakdown involves enzymes that can produce a toxic byproduct, so I'm cautious about anything that nudges that pathway, especially at high doses or with alcohol. For most folks taking normal doses this is very unlikely to matter. Still, run it by your pharmacist or doctor before adding lentinan, just to be safe.

From our pharmacist’s overview on the Acetaminophen, Aspirin page
“Acetaminophen with aspirin (Gemnisyn) is the kind of pain reliever people take without a second thought, which is exactly why its interaction list deserves genuine care. More than 200 of the 321 ingredients on file thin the blood in some way, and layered on top of aspirin that means a higher chance of bleeding, especially in the stomach.”

About Lentinan
What is Lentinan?
- Immune support
- Used alongside cancer treatment
- Anti-inflammatory
- General wellness/tonic
- Antimicrobial support
Lentinan is a beta-glucan polysaccharide from shiitake mushrooms that is studied mainly as an immune-modulating add-on to cancer chemotherapy, where it is given by injection in a few countries. Evidence for oral supplements is limited, and quality varies widely between products. Talk with your pharmacist or doctor before using it, especially if you have a health condition or are pregnant or breastfeeding.
