Esomeprazole, Naproxen falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Vimovo combines two medicines: esomeprazole (a stomach acid reducer) and naproxen (an anti-inflammatory pain reliever). The concern here comes from lab research showing that plumbagin, found in Ceylon Leadwort, may block a liver enzyme called CYP1A2. If that enzyme slows down, drugs it handles could build up a bit in your body.

Honestly, this one is mostly theoretical. The naproxen and esomeprazole in Vimovo aren't primarily cleared by CYP1A2, so a meaningful change isn't expected, and this hasn't been reported in real people. Still, since Ceylon Leadwort itself can irritate tissues and lacks good safety data, I'd run it by your pharmacist or doctor before combining them rather than guessing.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2C9 (CYP2C9) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2C9. Drugs that are metabolized by CYP2C9 include celecoxib (Celebrex), diclofenac (Voltaren), fluvastatin (Lescol), glipizide (Glucotrol), ibuprofen (Advil, Motrin), irbesartan (Avapro), losartan (Cozaar), phenytoin (Dilantin), piroxicam (Feldene), tamoxifen (Nolvadex), tolbutamide (Tolinase), torsemide (Demadex), and S-warfarin (Coumadin).

Vimovo combines naproxen (an anti-inflammatory pain reliever) with esomeprazole (a stomach-protecting acid blocker). The naproxen part is partly cleared by a liver enzyme called CYP2C9. A compound in Ceylon Leadwort, plumbagin, has been shown to slow that enzyme down, but only in lab dish studies, not in people. The idea is that if the enzyme works slower, naproxen could build up a bit and hang around longer.

In theory, higher naproxen levels might mean more of its usual side effects, like stomach upset, heartburn, fluid retention, or kidney strain. This is purely theoretical right now, so don't be alarmed. Still, since both naproxen and Ceylon Leadwort can irritate the gut, it's worth running this combo by your pharmacist or doctor before mixing them.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Vimovo combines two medicines: esomeprazole (an acid-reducing PPI) and naproxen (an anti-inflammatory pain reliever). The concern here is that plumbagin, a compound in Ceylon Leadwort, slowed down the CYP3A4 enzyme in lab dishes. Since this enzyme helps break down some of these medicines, the theory is that levels could creep up if the enzyme is blocked.

But here's the honest part: this is only test-tube data, never shown in real people, and naproxen actually relies more on a different enzyme (CYP2C9) than CYP3A4. So a big problem is unlikely. Still, if levels did rise, you might notice more naproxen side effects like stomach upset, heartburn, or a higher bleeding risk. Please check with your pharmacist or doctor before combining them.

ANTICOAGULANT/ANTIPLATELET DRUGS

Esomeprazole, Naproxen also falls under ANTICOAGULANT/ANTIPLATELET DRUGS.

There is some concern that Ceylon leadwort might potentiate the effects of anticoagulant and antiplatelet drugs and possibly increase the risk of bleeding. Ceylon leadwort root extract has been shown to decrease platelet adhesion and prolong bleeding in animals. However, this effect has not yet been demonstrated in humans. Until more is known, use cautiously in patients taking anticoagulant or antiplatelet drugs. Some of these drugs include aspirin, clopidogrel (Plavix), dalteparin (Fragmin), enoxaparin (Lovenox), heparin, ticlopidine (Ticlid), warfarin (Coumadin), and others.

Vimovo combines naproxen (an NSAID pain reliever) with esomeprazole (a stomach-acid blocker that's there to protect your gut). Naproxen already carries a known bleeding risk on its own, since it can thin the blood a bit and irritate the stomach lining. That's the piece that matters here.

In animal studies, Ceylon Leadwort root has slowed how platelets stick together and made bleeding last longer. Stack that on top of naproxen and the worry is additive bleeding risk, think easier bruising, nosebleeds, or stomach bleeding (dark stools, throwing up something that looks like coffee grounds). To be clear, this hasn't been shown in people yet, so it's a theoretical caution, not a proven danger. Still, I'd check with your pharmacist or doctor before pairing the two.
