Why this applies to Desipramine

Desipramine falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Red Clover interacts with.

Theoretically, red clover might increase levels of drugs metabolized by CYP1A2; however, a small clinical study found no effect.
In vitro evidence shows that red clover inhibits CYP1A2. However, a clinical study in healthy postmenopausal individuals shows that taking red clover capsules standardized to contain 60 mg isoflavones twice daily for 14 days does not affect the pharmacokinetics of caffeine, a CYP1A2 probe substrate.

Here's the gist for desipramine (Norpramin). Your body partly relies on an enzyme called CYP1A2 to clear this medication. In a test-tube setting, red clover's isoflavones can slow that enzyme down a little. If that happened in real life, desipramine levels could nudge upward, which in theory might mean more of its usual side effects like dry mouth, constipation, drowsiness, dizziness, or a faster heartbeat.

The reassuring part: this is mostly lab-dish evidence, and an actual human study using standardized red clover found no meaningful change in how a CYP1A2 drug was handled. So the real-world risk looks small and unlikely. Desipramine is still a drug worth respecting, so it's smart to mention any new supplement to your pharmacist or doctor.

Here's the gist with desipramine and red clover. In lab studies, red clover slightly slows down an enzyme called CYP2C19, which is one of the pathways your body uses to clear desipramine. If that pathway is even a little blocked, desipramine could build up a bit. In theory that might nudge up its usual side effects, things like dry mouth, constipation, drowsiness, dizziness when you stand up, or a faster heartbeat. Desipramine is an active medicine (not a prodrug), so more of it means stronger effects, not weaker.

That said, this is only a theoretical, test-tube finding, and it has never actually been reported in people. The effect looks weak and unlikely. Still, if you take desipramine and are thinking about red clover, it's worth a quick chat with your pharmacist or doctor first.

What Is Desipramine Used For?

Brand names: Norpramin, Desipramine, Pertofrane

Desipramine hydrochloride tablets — sold under the brand names Norpramin, Desipramine HCL, and Desipramine Hydrochloride — are FDA-approved for the treatment of depression in adults. Norpramin tablets and the generic formulations share this same approved indication.

Desipramine tablets are not approved for use in children. Adolescents and elderly patients may be prescribed it in certain circumstances, but at lower doses and under close supervision.
