1,3-dmba and Propranolol Hydrochloride

Here is the interaction we found between these two, with the evidence behind it.

Minor

CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES

Why this applies to Propranolol Hydrochloride

Propranolol Hydrochloride falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group 1,3-dmba interacts with.

Theoretically, 1,3-DMBA might modestly increase the levels of CYP2D6 substrates.
In vitro research shows that 1,3-DMBA weakly inhibits CYP2D6 enzymes.
Severity Minor Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the gist for propranolol. Your liver uses an enzyme called CYP2D6 to help clear this drug out of your system. In lab-dish studies, 1,3-DMBA weakly blocks that enzyme, so in theory propranolol could clear a little slower and build up slightly. That might mean a touch more of the usual beta-blocker effects, like a slower heart rate, lower blood pressure, tiredness, or feeling lightheaded.

Honestly though, the bigger worry is the stimulant itself. 1,3-DMBA is a banned, unsafe ingredient that can raise heart rate and blood pressure, which works directly against what propranolol is trying to do. The enzyme interaction is only theoretical, but the heart and stroke risks are real. Please avoid products with it and check with your doctor or pharmacist.

Likelihood how strong the evidence is that the interaction actually happens in people
  • LikelyWell-controlled human studies have demonstrated the likely existence of this interaction.
  • ProbableInteraction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports.
  • PossibleInteraction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
  • UnlikelyInteraction has been demonstrated in animal or in lab research but has been shown not to occur in humans.
Evidence the quality of the studies behind the rating (graded A–D)
  • AHigh-quality meta-analysis (quantitative systematic review).
  • BCase-control study.
  • CConsensus.
