Amitriptyline, Chlordiazepoxide falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Chanca Piedra interacts with.

Theoretically, chanca piedra might reduce the levels and clinical effects of CYP1A2 substrates.
In vitro research shows that chanca piedra extract increases CYP1A2 activity. Theoretically, chanca piedra might increase metabolism of CYP1A2 substrates and lower serum concentrations. This interaction has not been reported in humans.

Limbitrol DS combines amitriptyline (an antidepressant) with chlordiazepoxide (a calming benzodiazepine). The amitriptyline part is partly broken down by a liver enzyme called CYP1A2. In a lab setting, chanca piedra seemed to speed up that enzyme. In theory, that could clear amitriptyline a little faster and lower its level, which might make it work less well, so you could notice your mood symptoms or sleep slipping back.

Here's the honest part: this is only test-tube evidence and has never been reported in real people, so I wouldn't lose sleep over it. Neither of these medicines is something to stop or adjust on your own. If you'd like to try chanca piedra, just run it by your pharmacist or doctor first so they can keep an eye on how you're feeling.

In vitro research shows that chanca piedra extract inhibits CYP3A4. Theoretically, chanca piedra might increase the levels of CYP3A4 substrates. This interaction has not been reported in humans.

Limbitrol DS combines two medicines: amitriptyline (a tricyclic antidepressant) and chlordiazepoxide (a benzodiazepine). Both are partly broken down by a liver enzyme called CYP3A4. In lab tests, chanca piedra slows that enzyme down. The theory is that with the enzyme working slower, these two drugs could build up a bit more than usual in your body.

If that happened, you might feel the sedating side effects more strongly: extra drowsiness, dizziness, dry mouth, constipation, or feeling foggy. I want to be honest though, this is only a theoretical concern from test-tube research, and it has never actually been reported in people. Still, both of these medicines can already make you sleepy, so play it safe and check with your pharmacist or doctor before combining them.
