Entrectinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Chanca Piedra interacts with.

Theoretically, use of chanca piedra might increase the levels and clinical effects of CYP3A4 substrates.
In vitro research shows that chanca piedra extract inhibits CYP3A4. Theoretically, chanca piedra might increase the levels of CYP3A4 substrates. This interaction has not been reported in humans.

Here's the gist: your body relies on a liver enzyme called CYP3A4 to break down and clear entrectinib (Rozlytrek). In lab studies, chanca piedra appears to slow that same enzyme down. If that happened in a real person, entrectinib could build up a bit higher than expected, which might mean more of its known side effects, things like dizziness, fatigue, trouble with balance, swelling, changes in heart rhythm, or numbness and tingling.

I want to be honest about the strength of this, though. So far this is only test-tube data, and it has not been seen in actual patients. Still, because entrectinib is a cancer medicine where dosing really matters, I'd play it safe. Please check with your oncologist or pharmacist before adding chanca piedra rather than starting it on your own.

About Entrectinib

What Is Entrectinib Used For?

Brand name: Rozlytrek

Rozlytrek (entrectinib) is used to treat adult patients with ROS1-positive metastatic non-small cell lung cancer (NSCLC), as confirmed by an FDA-approved test. It is also approved for adult and pediatric patients older than 1 month of age who have solid tumors — any cancer type — that carry an NTRK gene fusion detected by an FDA-approved test, provided the tumor has no known acquired resistance mutation, is metastatic or cannot be surgically removed without severe harm, and has progressed after prior treatment or has no satisfactory alternative therapy.

The NTRK gene fusion approval in solid tumors was granted under accelerated approval, meaning it is based on tumor response rates. Continued approval may depend on results from confirmatory clinical trials.
