Amitriptyline, Perphenazine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Hordenine interacts with.

Hordenine weakly inhibits cytochrome P450 2D6 (CYP2D6) enzymes in vitro. Theoretically, hordenine might increase the levels of CYP2D6 substrates.
Some of drugs that are CYP2D6 substrates include amitriptyline (Elavil), clozapine (Clozaril), codeine, desipramine (Norpramin), donepezil (Aricept), fentanyl (Duragesic), flecainide (Tambocor), fluoxetine (Prozac), meperidine (Demerol), methadone (Dolophine), metoprolol (Lopressor, Toprol XL), olanzapine (Zyprexa), ondansetron (Zofran), tramadol (Ultram), trazodone (Desyrel), and others.

This combination pill pairs amitriptyline (a tricyclic antidepressant) with perphenazine (an antipsychotic), and both rely partly on a liver enzyme called CYP2D6 to get cleared from your body. Hordenine has been shown to weakly block that enzyme, but only in lab dish studies, not in real people. The theory is that if CYP2D6 is slowed, these two drugs could build up a bit, nudging up familiar side effects like drowsiness, dry mouth, constipation, dizziness on standing, or a faster heartbeat.

Honestly, the evidence here is thin and the effect is rated minor, so for most folks it's probably not a big deal. Still, since hordenine is also a stimulant and these drugs affect the heart and brain, I'd run it past your pharmacist or doctor before mixing them rather than guessing.
