Why this applies to Crizotinib
Crizotinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Chanca Piedra interacts with.
In vitro research shows that chanca piedra extract inhibits CYP3A4. Theoretically, chanca piedra might increase the levels of CYP3A4 substrates. This interaction has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist: your body relies on a liver enzyme called CYP3A4 to break down and clear crizotinib. Lab studies suggest chanca piedra can slow that same enzyme down. If it does that in real life, crizotinib could build up higher than intended, which may mean stronger side effects like nausea, vomiting, diarrhea, swelling, vision changes, fatigue, or changes in heart rhythm and liver enzymes.
I want to be honest about the strength of this, though. So far this is only seen in test-tube research, not in people taking chanca piedra and crizotinib together. Still, crizotinib is a serious cancer drug with a fairly narrow safety window, so I'd play it safe. Please check with your oncologist or pharmacist before adding this herb.

About Crizotinib
What Is Crizotinib Used For?
Brand name: Xalkori
Xalkori (crizotinib) treats adult patients with metastatic non-small cell lung cancer (NSCLC) whose tumors test positive for ALK or ROS1 gene changes, as confirmed by an FDA-approved test.
Xalkori is also used for pediatric patients (age 1 and older) and young adults with relapsed or refractory systemic ALK-positive anaplastic large cell lymphoma (ALCL), and for adult and pediatric patients (age 1 and older) with unresectable, recurrent, or refractory ALK-positive inflammatory myofibroblastic tumor (IMT). Xalkori's safety and effectiveness in older adults with relapsed or refractory ALK-positive ALCL have not been established.
