These two interact in 3 distinct ways. Each one is explained separately below.
Why Are There 3 Interactions?
Yerba Mate interacts with whole categories of medication. Aspirin, Caffeine, Orphenadrine falls into 3 of those categories, so it can interact with Yerba Mate in 3 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).
ANTICOAGULANT/ANTIPLATELET DRUGS
1 of 3Why this applies to Aspirin, Caffeine, Orphenadrine
Aspirin, Caffeine, Orphenadrine falls under ANTICOAGULANT/ANTIPLATELET DRUGS — the group Yerba Mate interacts with.
Yerba mate contains caffeine. Caffeine is reported to have antiplatelet activity. Theoretically, it might increase the risk of bleeding when used concomitantly with these agents; however, this interaction has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's a fun wrinkle with this combo: your medication already contains caffeine, and so does yerba mate. The interaction on file is mostly about the aspirin part. Aspirin thins the blood by blocking platelets, and caffeine is thought to add a small antiplatelet nudge of its own. On paper, stacking yerba mate on top could very slightly raise bleeding risk, like easier bruising, nosebleeds, or a cut that takes longer to stop.
Honestly though, this is theoretical and based on lab work, not real patient reports, so I wouldn't lose sleep over it. The bigger practical issue is doubling up on caffeine, which can leave you jittery, racing-hearted, or unable to sleep. If you drink mate daily, just mention it to your pharmacist or doctor.

STIMULANT DRUGS
2 of 3Aspirin, Caffeine, Orphenadrine also falls under STIMULANT DRUGS.
Yerba mate contains caffeine. Due to the CNS stimulant effects of the caffeine, concomitant use can increase the risk of adverse effects.
Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports..
Here's the simple version: this combination product already has caffeine built right in, alongside aspirin and the muscle relaxer orphenadrine. Yerba mate brings its own caffeine to the table, so drinking it adds even more stimulant on top of what you're already taking. That stacking is the whole concern here.
When caffeine piles up, you may notice the familiar overstimulation signs: a racing or pounding heart, jitteriness, trouble sleeping, an upset stomach, or feeling anxious and on edge. The orphenadrine piece can also dry you out a bit. None of this is usually dangerous, and it's based mostly on expert reasoning rather than hard studies. Still, if you're sensitive to caffeine or notice these effects, it's worth chatting with your pharmacist or doctor about how much mate is sensible for you.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
3 of 3Aspirin, Caffeine, Orphenadrine also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.
In vitro research shows that yerba mate extract inhibits CYP3A4 enzymes. Theoretically, taking yerba mate may increase levels and adverse effects of CYP3A4 substrates.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with this combo pill (which packs aspirin, caffeine, and the muscle relaxant orphenadrine). Lab studies hint that yerba mate may gently slow down a liver enzyme called CYP3A4, which helps clear orphenadrine. If that happens, the orphenadrine could linger a bit longer and feel stronger, meaning more drowsiness, dry mouth, blurry vision, or dizziness.
Just as important, your mate and this medicine both carry caffeine. Stacking them can leave you jittery, racing-hearted, or unable to sleep. Honestly, the evidence here is theoretical and only from test-tube work, so don't panic. Still, if you drink mate daily, mention it to your pharmacist or doctor so they can keep an eye on how you feel.

About Yerba Mate
What is Yerba Mate?
- Energy and alertness
- Weight loss
- Mental focus
- Digestive support
- Antioxidant support
Yerba mate is a caffeine-containing herbal beverage from South America that is widely enjoyed for its stimulating, coffee-like effects. While it is rich in antioxidants and is being studied for weight, blood sugar, and cholesterol, strong human evidence for most health claims is limited. Drinking very hot mate over many years has been linked to higher risk of certain cancers, so enjoy it warm and in moderation.
