Hemp and Vamorolone

Here is the interaction we found between these two, with the evidence behind it.

Minor

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

Why this applies to Vamorolone

Vamorolone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Hemp interacts with.

Theoretically, hemp might decrease the levels and clinical effects of CYP3A4 substrates.
In vitro research shows that hemp induces CYP3A4 enzymes.
Severity Minor Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the short version for anyone on Vamorolone (Agamree). Your body clears this steroid using a liver enzyme called CYP3A4. Some early lab research suggests hemp may speed that enzyme up, which in theory could clear Vamorolone a little faster and make it slightly less effective at controlling muscle inflammation in Duchenne muscular dystrophy.

I want to be honest with you though: this is graded as a minor, theoretical concern based only on test-tube studies, not real people. Eating hemp seeds or hemp oil as food is very unlikely to cause a problem. If you're using concentrated hemp supplements daily, just mention it to your pharmacist or your child's neurologist so they can keep an eye on how well the medication is working. Don't change anything on your own.

Likelihood how strong the evidence is that the interaction actually happens in people
  • LikelyWell-controlled human studies have demonstrated the likely existence of this interaction.
  • ProbableInteraction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports.
  • PossibleInteraction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
  • UnlikelyInteraction has been demonstrated in animal or in lab research but has been shown not to occur in humans.
Evidence the quality of the studies behind the rating (graded A–D)
  • AHigh-quality meta-analysis (quantitative systematic review).
  • BCase-control study.

From our pharmacist’s overview on the Vamorolone page

“Vamorolone (Agamree), a steroid for Duchenne muscular dystrophy, often taken by children, deserves real caution with supplements because nearly everything on this list acts on one liver enzyme, CYP3A4, which controls how much of the drug stays in the body. Herbs that block the enzyme can raise vamorolone levels and amplify steroid side…”

About Hemp

What is Hemp?

Hemp
  • Plant-based protein source
  • Heart health support
  • Skin and hair health
  • Reducing inflammation
  • General nutrition (omega fatty acids)

Hemp seeds and hemp seed oil are nutritious foods rich in protein, fiber, and healthy omega-3 and omega-6 fatty acids, and they are generally safe for most people as part of the diet. While the nutrition is real, strong human evidence that hemp supplements treat specific medical conditions is limited. Talk with your pharmacist or doctor before using hemp products for a health concern, especially if you are pregnant or breastfeeding.
