Why this applies to Trazodone

Trazodone falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Lentinan interacts with.

Laboratory research suggests that lentinan might increase the activity of cytochrome P450 2D6 (CYP2D6). So far, this interaction has not been reported in humans. However, watch for a decrease in the levels of drugs metabolized by CYP2D6 in patients taking lentinan.
Some drugs metabolized by CYP2D6 include tricyclic antidepressants such as imipramine (Tofranil) and amitriptyline (Elavil); antipsychotics such as haloperidol (Haldol), risperidone (Risperdal), and chlorpromazine (Thorazine); beta-blockers such as propranolol (Inderal), metoprolol (Lopressor, Toprol XL), and carvedilol (Coreg); tamoxifen (Nolvadex); and others.

Here's the short version for trazodone and lentinan. Your liver uses an enzyme called CYP2D6 to help clear trazodone from your body. Lab studies hint that lentinan might speed up that enzyme, which could mean your body breaks trazodone down a little faster. If that happened, you might notice trazodone working less well, with depression symptoms or sleep troubles creeping back.

The important part: this has only been seen in test tube and animal research, never confirmed in real people, so it's more theoretical than proven. I wouldn't lose sleep over it. Still, if you decide to try a lentinan supplement, mention it to your pharmacist or doctor first so they can keep an eye on how well your trazodone is working.

Laboratory research suggests that lentinan might increase the activity of cytochrome P450 3A4 (CYP3A4). So far, this interaction has not been reported in humans. However, watch for a decrease in the levels of drugs metabolized by CYP3A4 in patients taking lentinan.
Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others.

Here's the gist for trazodone and lentinan. Your liver uses an enzyme called CYP3A4 to break down trazodone. Lab studies hint that lentinan might speed up that enzyme, which could clear trazodone out of your system a bit faster. Trazodone is the active drug (not a prodrug that needs activating), so faster clearance would mean less of it in your blood, and potentially a weaker effect.

If that happened, you might notice your depression or sleep not being as well controlled as usual. That said, this is only seen in lab and animal research, not in real people, so I wouldn't lose sleep over it. Don't change anything on your own. Just give your pharmacist or doctor a heads-up if you want to add lentinan.
