Coffee + Protriptyline
Protriptyline brand name: Vivactil.
Coffee has a minor interaction with Protriptyline. Theoretically, TCAs might bind with coffee constituents when taken at the same time.

Here is the interaction we found between these two, with the evidence behind it.
TRICYCLIC ANTIDEPRESSANTS (TCAs)
Why this applies to Protriptyline
Protriptyline falls under TRICYCLIC ANTIDEPRESSANTS (TCAs) — the group Coffee interacts with.
In vitro research suggests that the tannins in coffee might cause precipitation of TCAs. However, this effect has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with coffee and protriptyline: in a lab setting, the tannins in coffee can clump together with tricyclic antidepressants like this one. The worry is that if they bind up in your stomach, your body might absorb a bit less of the medicine, which could make it work less well over time. That might look like your depression not lifting the way it should.
The reassuring part? This has only been seen in test tubes, never actually reported in people. So it's theoretical and rated minor. If you want to be cautious, separating your coffee from your dose by an hour or two is an easy habit. Don't change anything on your own, but feel free to run it by your pharmacist or doctor.

Clinically significant. Avoid the combination, or use only under close professional supervision.
Usually worth avoiding or managing — may need monitoring, a dose change, or timing apart.
Generally not serious. Be aware of it and mention it to your pharmacist.
- LikelyWell-controlled human studies have demonstrated the likely existence of this interaction.
- ProbableInteraction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports.
- PossibleInteraction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
- UnlikelyInteraction has been demonstrated in animal or in lab research but has been shown not to occur in humans.

From our pharmacist’s overview on the Protriptyline page
“Protriptyline (Vivactil) is an older tricyclic antidepressant, and I read its supplement list with genuine care because so much of it piles onto what the drug already does. Four of the 93 ingredients on file rate major, and those deserve your attention before anything else.”

About Coffee
What is Coffee?
- Alertness and reducing fatigue
- Mental focus and concentration
- Athletic performance
- Headache relief
- Weight management
Coffee is a widely consumed beverage made from roasted coffee beans, valued mainly for its caffeine, which boosts alertness and energy. For most healthy adults, moderate coffee intake is generally safe and may have some health benefits, but too much can cause jitteriness, poor sleep, and a faster heartbeat. People who are pregnant, breastfeeding, or sensitive to caffeine should limit their intake and check with a doctor or pharmacist.

Protriptyline Is Linked to Lower Levels of 3 Nutrients
Natural Medicines records Protriptyline as a medicine that can lower these. This is not an interaction with Coffee — it is a separate effect of the medicine, and it is a reason some people are advised to supplement rather than to avoid one:
