Theoretically, theanine may compete with glutamate and/or increase plasma gamma-aminobutyric acid (GABA) levels, which could cause CNS depression. In one clinical study, some subjects taking oral theanine reported drowsiness.

Here's the simple version: your Tussend already has two ingredients that can make you sleepy, the antihistamine (chlorpheniramine) and the opioid (hydrocodone). Theanine is a calming amino acid from tea, and on paper it may nudge up your brain's relaxing GABA signals. So the worry is just a little extra drowsiness when you stack them together.
I want to be honest about how soft this concern is. It's theoretical, the risk is rated minor and unlikely, and the main thing a few people noticed with theanine was mild sleepiness. Still, watch for feeling foggy, extra tired, or slowed down, and don't drive until you know how you react. Run it past your pharmacist or doctor before combining them.

Theanine may nudge serotonin levels in the brain, though honestly the research pulls in both directions, with some studies showing it goes up and others showing it goes down. There's no real-world report of theanine triggering serotonin-related problems, so this is a theoretical, low-priority flag.
Practically speaking, none of the three ingredients in Tussend is a heavy serotonin drug, so I wouldn't expect trouble. I'd pay more attention to overlapping calming effects: theanine plus the antihistamine and hydrocodone could add up to extra drowsiness.
