Celecoxib, Tramadol falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Yohimbe interacts with.

Theoretically, yohimbe might increase the levels and adverse effects of CYP2D6 substrates.
In vitro research suggests that yohimbine, a constituent of yohimbe bark, inhibits CYP2D6 enzyme activity.

Seglentis combines two medicines, and the tramadol part is the one to watch here. Tramadol relies on a liver enzyme called CYP2D6 to turn into its stronger pain-relieving form. Yohimbine, the active piece of yohimbe, has shown in lab studies that it can slow this same enzyme down. In theory, that could throw tramadol's effect off balance, either dulling some of the pain relief or letting more of the parent drug linger, which may add to side effects like nausea, dizziness, drowsiness, or constipation.

Honestly, this is based on test-tube research only, so it's more of a question mark than a proven problem. The bigger concern for many people is yohimbe itself, since it can spike blood pressure and heart rate. Please check with your pharmacist or doctor before combining them.

CYP2D6 inhibitors may increase the levels and adverse effects of yohimbine, a constituent of yohimbe.
In vitro and clinical research shows that the yohimbe bark constituent, yohimbine, is metabolized by CYP2D6 isoenzymes. Paroxetine, a cytochrome P450 (CYP) 2D6 inhibitor, increases the maximum serum concentration of yohimbine and reduces the clearance of yohimbine compared to yohimbine alone in patients who are extensive CYP2D6 metabolizers..

Here's the key thing with this combo: the Seglentis product contains tramadol, and tramadol can block the liver enzyme CYP2D6. That same enzyme is what your body uses to break down yohimbine, the active compound in yohimbe. So when you take them together, the yohimbine isn't cleared as quickly and can build up to higher levels than expected.

Why does that matter? Yohimbine at higher levels is known to push up blood pressure, cause a racing or pounding heart, anxiety, jitteriness, and trouble sleeping. Tramadol itself can also raise heart rate and stir up the nervous system, so stacking the two could amplify those effects. This is rated a moderate, likely interaction. I'd hold off on combining them and check with your pharmacist or doctor first.

Seglentis pairs two medicines: celecoxib (an anti-inflammatory for pain) and tramadol (an opioid-type pain reliever that your liver has to switch on to work fully). Both pass through a liver enzyme called CYP3A4. In lab dish studies, yohimbe seemed to speed that enzyme up, which could in theory clear these drugs a bit faster and make your pain relief weaker.

Honestly, this is very weak evidence (test tube only), so I would not lose sleep over it. The bigger worry with yohimbe is its own punch: it can raise blood pressure and speed up your heart, which is not something you want alongside pain medicine. Please run it by your pharmacist or doctor before combining them.

Yohimbe is a West African tree bark that contains yohimbine, a compound mainly promoted for erectile dysfunction and as an aphrodisiac. A prescription form of yohimbine has some evidence for erectile dysfunction, but over-the-counter yohimbe supplements vary widely in content and can cause serious side effects like high blood pressure and fast heart rate. Because of these safety concerns, talk to your pharmacist or doctor before using it.
