Here's the situation with these two together. Your body relies on a liver enzyme called CYP2D6 to break down and clear desipramine. Lab studies hint that yohimbine, the active piece of yohimbe, may slow that enzyme down. If that happens in real life, desipramine could build up in your system and hit harder than intended, bringing on side effects like a racing or pounding heartbeat, dizziness, dry mouth, constipation, or trouble urinating.

I want to be honest that this is only based on test-tube research, so we don't really know how much it matters in people. Still, yohimbe on its own can spike blood pressure and heart rate, which overlaps with desipramine's own heart effects. Please check with your pharmacist or doctor before combining them.

CYP2D6 inhibitors may increase the levels and adverse effects of yohimbine, a constituent of yohimbe.
In vitro and clinical research shows that the yohimbe bark constituent, yohimbine, is metabolized by CYP2D6 isoenzymes. Paroxetine, a cytochrome P450 (CYP) 2D6 inhibitor, increases the maximum serum concentration of yohimbine and reduces the clearance of yohimbine compared to yohimbine alone in patients who are extensive CYP2D6 metabolizers..

Here's the key thing to know if you're on desipramine (Norpramin) and thinking about a yohimbe supplement. Your body relies on a liver enzyme called CYP2D6 to clear out yohimbine, the active piece of yohimbe. Desipramine can slow that same enzyme down, so the yohimbine isn't broken apart as quickly. That means it can build up to higher levels than expected from the dose on the label.

Practically, that raises the odds of yohimbe's stimulant-type side effects: a racing or pounding heartbeat, spiking blood pressure, jitteriness, and anxiety. Since desipramine itself can already nudge your heart rate and blood pressure, stacking the two isn't ideal. This is probable rather than just theoretical, so please check with your pharmacist or doctor before combining them. Don't change your desipramine on your own.

Desipramine also falls under TRICYCLIC ANTIDEPRESSANTS (TCAs).

Theoretically, taking yohimbe with TCAs can increase adverse effects.
A small clinical study in patients taking TCAs for at least 4 weeks shows that receiving doses of intravenous yohimbine 2.5-20 mg daily for up to 7 days precipitates severe anxiety, agitation, and tremor. The effects of yohimbe bark itself are unclear; oral yohimbe bark contains 0.6% to 1.38% yohimbine, but it is unclear how much is absorbed.

Here's the gist for desipramine: this antidepressant already nudges up your body's stress chemicals (like norepinephrine) to help lift mood. Yohimbe's active piece, yohimbine, works in a similar direction by flipping on those same adrenaline-type signals. Stack the two and you can get too much of that effect at once.

In one small study, people on tricyclics who were given yohimbine ran into severe anxiety, jitteriness, agitation, and tremor. You might also notice a racing heart or a spike in blood pressure. That said, this is mostly theoretical, and we don't know how much yohimbine actually gets absorbed from over-the-counter bark, which varies a lot. Still, since both push the same buttons, please check with your pharmacist or doctor before pairing them.

Here's the gist for you. Lab studies suggest yohimbe may speed up an enzyme called CYP1A2, which is one of the pathways your body uses to clear desipramine (Norpramin). If that happens, your desipramine levels could drift a little lower, which might make it work less well for your depression over time.

I want to keep this in perspective. This is rated minor and is based only on test-tube research, not real patients, so it's more theoretical than proven. The bigger worry honestly is yohimbe itself, since it can raise blood pressure and heart rate, and tricyclics like desipramine can also affect your heart. Yohimbe is a West African tree bark that contains yohimbine, a compound mainly promoted for erectile dysfunction and as an aphrodisiac. A prescription form of yohimbine has some evidence for erectile dysfunction, but over-the-counter yohimbe supplements vary widely in content and can cause serious side effects like high blood pressure and fast heart rate. Because of these safety concerns, talk to your pharmacist or doctor before using it.

What Is Desipramine Used For?

Brand names: Norpramin, Desipramine, Pertofrane

Desipramine hydrochloride tablets — sold under the brand names Norpramin, Desipramine HCL, and Desipramine Hydrochloride — are FDA-approved for the treatment of depression in adults. Norpramin tablets and the generic formulations share this same approved indication.

Desipramine tablets are not approved for use in children. Adolescents and elderly patients may be prescribed it in certain circumstances, but at lower doses and under close supervision.
