Asciminib falls under CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS — the group Yohimbe interacts with.

Theoretically, CYP3A4 inhibitors might increase the levels and adverse effects of yohimbine, a constituent of yohimbe bark.
In vitro and clinical research shows that the yohimbe bark constituent, yohimbine, is metabolized by CYP3A4 enzymes. Theoretically, drugs that inhibit CYP3A4 might increase the levels and adverse effects of yohimbine.

Here's the thing to know about this pairing: your leukemia medicine Scemblix (asciminib) can mildly slow down a liver enzyme called CYP3A4, and that same enzyme is what your body uses to clear yohimbine, the active ingredient in yohimbe. So in theory, taking yohimbe alongside asciminib could let yohimbine build up a bit more than usual, which may make yohimbe's own side effects stronger.

What does that look like in real life? Mostly yohimbe-type effects, like a racing or pounding heartbeat, jitteriness, anxiety, and spikes in blood pressure. It does not make your asciminib weaker or stronger. Keep in mind this is only a theoretical concern based on how the two are processed, not something proven in studies. Still, since you're managing CML, please run yohimbe by your pharmacist or oncologist before trying it.

Yohimbe is a West African tree bark that contains yohimbine, a compound mainly promoted for erectile dysfunction and as an aphrodisiac. A prescription form of yohimbine has some evidence for erectile dysfunction, but over-the-counter yohimbe supplements vary widely in content and can cause serious side effects like high blood pressure and fast heart rate. Because of these safety concerns, talk to your pharmacist or doctor before using it.

What Is Asciminib Used For?

Brand name: Scemblix

Scemblix (asciminib) is used to treat adult patients with Philadelphia chromosome-positive chronic myeloid leukemia in chronic phase (Ph+ CML-CP) — a type of blood cancer driven by an abnormal protein called BCR-ABL1. It covers three groups: people newly diagnosed with Ph+ CML-CP, people who have already been treated for Ph+ CML-CP, and people whose cancer carries a specific mutation called T315I that makes it harder to treat.

The use in newly diagnosed Ph+ CML-CP was approved under accelerated approval based on early response data. Continued approval for that specific use may depend on results from confirmatory trials confirming long-term clinical benefit.
