Theanine + Amitriptyline, Perphenazine
Amitriptyline, Perphenazine brand names: Triavil, Etrafon, Etrafon-A, Etrafon-Forte.
Amitriptyline, Perphenazine falls into 2 drug categories that Theanine can interact with; the most serious is rated minor. Theoretically, theanine might have additive sedative effects when used in conjunction with CNS depressants.

These two interact in 2 distinct ways. Each one is explained separately below.
Why Are There 2 Interactions?
Theanine interacts with whole categories of medication. Amitriptyline, Perphenazine falls into 2 of those categories, so it can interact with Theanine in 2 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Minor).
CNS DEPRESSANTS
1 of 2Why this applies to Amitriptyline, Perphenazine
Amitriptyline, Perphenazine falls under CNS DEPRESSANTS — the group Theanine interacts with.
Theoretically, theanine may compete with glutamate and/or increase plasma gamma-aminobutyric acid (GABA) levels, which could cause CNS depression. In one clinical study, some subjects taking oral theanine reported drowsiness.
Interaction has been demonstrated in animal or in lab research but has been shown not to occur in humans..
Triavil combines amitriptyline (a tricyclic antidepressant) with perphenazine (an antipsychotic), and both can already make you feel sleepy or foggy on their own. Theanine, the calming amino acid from tea, may gently nudge your brain toward relaxation by influencing GABA. So the worry here is simple: stacking a calming supplement on top of an already-sedating combo pill could add up to extra drowsiness, grogginess, or feeling a bit slowed down.
That said, this is a theoretical concern based on how these things work, not strong real-world proof. It's rated minor and unlikely. If you do try theanine, just watch for drowsiness or dizziness, especially when driving, and run it by your pharmacist or doctor first.

SEROTONERGIC DRUGS
2 of 2Amitriptyline, Perphenazine also falls under SEROTONERGIC DRUGS.
Interaction has been demonstrated in animal or in lab research but has been shown not to occur in humans..
This combination pill pairs amitriptyline (an older antidepressant) with perphenazine (an antipsychotic). The amitriptyline side gently nudges serotonin levels in the brain, and theanine may have a small influence on serotonin too. On paper, putting two things that touch serotonin together raises the question of whether they could stack up.
Here's the honest picture: the research on theanine and serotonin actually points in both directions, and there are no reported cases of theanine triggering serotonin syndrome. So this is a very weak, theoretical concern, not something I'd expect to cause trouble. Just keep an eye out for things like extra drowsiness, dry mouth, or feeling restless, and check with your pharmacist or doctor before adding theanine, especially since this medicine already carries sedating effects.
What These Ratings Mean
Every interaction above is graded three ways — here’s the key to each label.
Clinically significant. Avoid the combination, or use only under close professional supervision.
Usually worth avoiding or managing — may need monitoring, a dose change, or timing apart.
Generally not serious. Be aware of it and mention it to your pharmacist.

From our pharmacist’s overview on the Amitriptyline, Perphenazine page
“Alcohol is the risk I'd flag first for anyone taking amitriptyline with perphenazine (Triavil, Etrafon). Beer and wine both carry major ratings here, and 78 of the 311 ingredients on file deepen the drowsiness this antidepressant and antipsychotic combination already causes, sometimes to the point of slowed breathing.”

About Theanine
What is Theanine?
- Anxiety and stress
- Relaxation and calm
- Sleep support
- Focus and cognitive function
Theanine (usually L-theanine) is an amino acid found naturally in tea leaves that many people take to feel calmer and less stressed without strong drowsiness. Early research suggests it may help with relaxation and stress, but evidence for most uses is still limited. It appears generally well tolerated for short-term use, but check with your pharmacist or doctor before starting it.
