Capmatinib Hydrochloride falls under HEPATOTOXIC DRUGS — the group Kava interacts with.

Theoretically, using kava with hepatotoxic drugs might increase the risk of liver damage.
Kava has been linked with over 100 cases of hepatotoxicity. Most cases occur with excessive and prolonged use. There is some concern that kava can adversely affect the liver, especially when used in combination with hepatotoxic drugs.

Here's the concern with pairing kava and capmatinib (Tabrecta). This isn't about kava changing how the drug works in your body. Instead, both can be hard on your liver. Capmatinib already carries a known risk of liver problems, and your oncology team likely checks your liver enzymes with regular blood tests while you're on it. Kava has its own track record of rare but serious liver injury, so stacking the two could add up.

To be honest, this interaction is based on theory and scattered case reports, not strong studies. Still, with a cancer drug that's already watched for liver strain, I'd play it safe. Please don't change anything on your own, just talk with your pharmacist or oncologist before using kava.

Capmatinib Hydrochloride also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.

It is unclear if kava inhibits CYP3AA; research is conflicting.
Although in vitro research suggests that kava inhibits CYP3A4, more robust clinical evidence shows that kava has no effect on CYP3A4. In a clinical study in healthy volunteers, taking kava 1000 mg twice daily (containing a daily dose of 138 mg kavalactones) for 28 days had no effect on CYP3A4 activity.

Here's the gist for capmatinib (Tabrecta) and kava. Your body clears capmatinib partly through a liver enzyme called CYP3A4. The theory was that kava might slow this enzyme, which could let capmatinib build up and bring on more of its usual side effects like nausea, swelling, tiredness, or higher liver enzymes. But the actual evidence doesn't support much worry here. In a study of healthy people, a real-world kava dose taken daily for a month had no measurable effect on CYP3A4.

So this interaction is rated minor and unlikely. That said, kava itself carries a rare risk of liver injury, and capmatinib can also stress the liver, so I'd still run kava by your oncologist or pharmacist before using it.

Tabrecta (capmatinib) is used to treat adults with metastatic non-small cell lung cancer (NSCLC) — lung cancer that has spread to other parts of the body. It is only for patients whose tumors have a specific genetic change called a MET exon 14 skipping mutation, which must be confirmed by an FDA-approved test before starting treatment.
