Sotorasib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Yohimbe interacts with.

Theoretically, yohimbe might decrease the levels and clinical effects of CYP3A4 substrates.
In vitro research shows that yohimbe extract induces CYP3A4 enzymes.

Here's the basic idea with Lumakras (sotorasib) and yohimbe. Your liver uses an enzyme called CYP3A4 to handle sotorasib, and lab studies suggest yohimbe might speed that enzyme up a bit. If it sped things up in real life, your body could clear sotorasib a little faster and leave less of it working against your cancer. Sotorasib is not a prodrug, so faster clearance would mean weaker effect, not stronger.

I want to be honest, though: this comes only from test-tube research, not from people, so it's mostly theoretical and rated as minor. Still, with a cancer medicine you don't want anything quietly reducing, so please run yohimbe by your oncology team before starting it.

Yohimbe is a West African tree bark that contains yohimbine, a compound mainly promoted for erectile dysfunction and as an aphrodisiac. A prescription form of yohimbine has some evidence for erectile dysfunction, but over-the-counter yohimbe supplements vary widely in content and can cause serious side effects like high blood pressure and fast heart rate. Because of these safety concerns, talk to your pharmacist or doctor before using it.

About Sotorasib

What Is Sotorasib Used For?

Brand name: Lumakras

Lumakras is used to treat adults with KRAS G12C-mutated locally advanced or metastatic non-small cell lung cancer (NSCLC) who have already received at least one prior cancer treatment. This use was approved under the FDA's accelerated approval pathway based on tumor response rates, and continued approval may depend on results from confirmatory trials.

Lumakras is also used in combination with panitumumab to treat adults with KRAS G12C-mutated metastatic colorectal cancer (mCRC) who have previously received certain chemotherapy regimens. In both cases, patients must first be confirmed to carry the KRAS G12C mutation using an FDA-approved test.
