Theoretically, CYP3A4 inhibitors might increase the levels and adverse effects of yohimbine, a constituent of yohimbe bark.
In vitro and clinical research shows that the yohimbe bark constituent, yohimbine, is metabolized by CYP3A4 enzymes. Theoretically, drugs that inhibit CYP3A4 might increase the levels and adverse effects of yohimbine.

Here's the thing to know about pairing yohimbe with Zileuton (Zyflo). Your body relies on an enzyme called CYP3A4 to help break down yohimbine, the active piece of yohimbe bark. Zileuton can slow that enzyme down a bit, so in theory the yohimbine could hang around longer and build up. That means you might feel more of yohimbe's stimulant-type effects, things like a racing heartbeat, jitteriness, anxiety, or a jump in blood pressure.

I'll be straight with you: this concern is theoretical, based on how the two are processed rather than on real-world reports of trouble. Still, since yohimbe can already strain your heart, it's worth a quick chat with your pharmacist or doctor before combining them.

Here's the basic idea with yohimbe and zileuton (Zyflo). Zileuton is your daily asthma controller, broken down in part by a liver enzyme called CYP1A2. Lab studies suggest yohimbe may speed that enzyme up, which could clear zileuton out faster and leave a bit less of it working in your body.

If that happened, zileuton might do a weaker job keeping your airways calm, so you could notice more wheezing, coughing, or chest tightness over time. I want to be honest, though: this is only seen in test-tube research, not in real people, so it's mostly theoretical and rated minor. Zileuton is not a prodrug, so it doesn't flip the other way. Before adding yohimbe, run it past your pharmacist or doctor, especially since yohimbe itself can raise blood pressure and heart rate.

Here's the gist for these two together. Lab studies suggest yohimbe may speed up a liver enzyme called CYP3A4, which is one of the enzymes that helps clear zileuton. If that enzyme runs faster, your body could break down zileuton a little quicker, leaving slightly less of it around to do its job. In practice, that might mean your asthma control isn't quite as steady, with more wheezing, coughing, or chest tightness sneaking back in.

That said, I want to keep this in perspective. This is a theoretical concern based only on test-tube research, and it's rated as minor, so for most people it's unlikely to cause real trouble. Zileuton isn't a prodrug, so it isn't flipped the other way. Still, since yohimbe itself can raise blood pressure and heart rate, I'd run it by your pharmacist or doctor before combining them.

Yohimbe is a West African tree bark that contains yohimbine, a compound mainly promoted for erectile dysfunction and as an aphrodisiac. A prescription form of yohimbine has some evidence for erectile dysfunction, but over-the-counter yohimbe supplements vary widely in content and can cause serious side effects like high blood pressure and fast heart rate. Because of these safety concerns, talk to your pharmacist or doctor before using it.

About Zileuton

What Is Zileuton Used For?

Brand names: Zyflo, Zyflo CR

Zileuton is used for the ongoing prevention and treatment of asthma in adults and adolescents aged 12 and older. It is a maintenance medicine — meaning it is taken regularly to keep asthma under control, not to stop an asthma attack once it has started.

Zyflo (the tablet form) and zileuton extended-release tablets are both approved for this same purpose. Neither form should be used to treat a sudden asthma attack. However, both can be continued during a flare-up of asthma while you also use your rescue inhaler.
