Crizotinib falls under CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS — the group Yohimbe interacts with.

Theoretically, CYP3A4 inhibitors might increase the levels and adverse effects of yohimbine, a constituent of yohimbe bark.
In vitro and clinical research shows that the yohimbe bark constituent, yohimbine, is metabolized by CYP3A4 enzymes. Theoretically, drugs that inhibit CYP3A4 might increase the levels and adverse effects of yohimbine.

Here the spotlight is really on the yohimbe, not your cancer medicine. Crizotinib (Xalkori) acts as a blocker of the CYP3A4 enzyme, the same enzyme that breaks down yohimbine, the active compound in yohimbe bark. When that enzyme is slowed down, yohimbine can build up in your body and hit harder than expected.

In plain terms, this could mean more of yohimbe's own troublesome effects: a racing or pounding heart, spikes in blood pressure, anxiety, jitteriness, or trouble sleeping. That is a real concern when you are already managing cancer treatment. Keep in mind this is a theoretical interaction based on how the body processes these substances, not strong human studies. Still, given yohimbe's safety record, please check with your pharmacist or oncologist before using it.

Here's the gist for Xalkori (crizotinib): your body relies on a liver enzyme called CYP3A4 to break this cancer drug down. Lab studies suggest yohimbe may speed that enzyme up a bit. In theory, a faster cleanup could lower the amount of crizotinib in your blood, which matters because you want steady, reliable levels to keep working against your lung cancer or tumor.

I want to be honest about how soft this concern is. It comes only from test-tube research, not from real patients, so it's more of a 'worth knowing' than a 'sound the alarm.' Crizotinib is too important to leave to guesswork, though. Please loop in your oncologist or pharmacist before adding yohimbe, especially since this supplement can also raise blood pressure and heart rate on its own.

Yohimbe is a West African tree bark that contains yohimbine, a compound mainly promoted for erectile dysfunction and as an aphrodisiac. A prescription form of yohimbine has some evidence for erectile dysfunction, but over-the-counter yohimbe supplements vary widely in content and can cause serious side effects like high blood pressure and fast heart rate. Because of these safety concerns, talk to your pharmacist or doctor before using it.

About Crizotinib

What Is Crizotinib Used For?

Brand name: Xalkori

Xalkori (crizotinib) treats adult patients with metastatic non-small cell lung cancer (NSCLC) whose tumors test positive for ALK or ROS1 gene changes, as confirmed by an FDA-approved test.

Xalkori is also used for pediatric patients (age 1 and older) and young adults with relapsed or refractory systemic ALK-positive anaplastic large cell lymphoma (ALCL), and for adult and pediatric patients (age 1 and older) with unresectable, recurrent, or refractory ALK-positive inflammatory myofibroblastic tumor (IMT). Xalkori's safety and effectiveness in older adults with relapsed or refractory ALK-positive ALCL have not been established.
